Phagocytosis and killing assays were determined using microscopy and viable colony plate counts. The results indicated a highly efficient phagocytosis effect of J774.1 cells against both species of Penicillium. There was no difference between the percentages of cells phagocytosed in both species. The phagocytic indices of P. marneffei at 60, 120, and 240 minutes of infection were significantly higher than those of P. citrinum. However, in the early stages of phagocytosis, the percentage killing of P. marneffei was significantly lower than that observed in P. citrinum. Conidia of P. marneffei were more susceptible to being phagocytosed but also more resistant to killing by macrophages than those of non-pathogenic Penicillium. The mechanism of intracellular survival or resistance to killing by macrophages needs further investigation. (J Infect Dis Antimicrob Agents 2008;25:81-9.)

Morphogenetic circuitry regulating growth and development in the dimorphic pathogen Penicillium marneffei.

Penicillium marneffei is an emerging human-pathogenic fungus endemic to Southeast Asia. Like a number of other fungal pathogens, P. marneffei exhibits temperature-dependent dimorphic growth and grows in two distinct cellular morphologies, hyphae at 25°C and yeast cells at 37°C. Hyphae can differentiate to produce the infectious agents, asexual spores (conidia), which are inhaled into the host l...

Caenorhabditis elegans: A Simple Nematode Infection Model for Penicillium marneffei

Penicillium marneffei, one of the most important thermal dimorphic fungi, is a severe threat to the life of immunocompromised patients. However, the pathogenic mechanisms of P. marneffei remain largely unknown. In this work, we developed a model host by using nematode Caenorhabditis elegans to investigate the virulence of P. marneffei. Using two P. marneffei clinical isolate strains 570 and 486...

Functional Analysis of atfA Gene to Stress Response in Pathogenic Thermal Dimorphic Fungus Penicillium marneffei

Penicillium marneffei, the pathogenic thermal dimorphic fungus is a causative agent of a fatal systemic disease, penicilliosis marneffei, in immunocompromised patients especially HIV patients. For growth and survival, this fungus has to adapt to environmental stresses outside and inside host cells and this adaptation requires stress signaling pathways and regulation of gene expression under var...

Effects of iron on extracellular and intracellular growth of Penicillium marneffei.

Killing of intracellular Penicillium marneffei conidia is demonstrated in gamma interferon-lipopolysaccharide-activated human THP1 and mouse J774 cells. Iron overload significantly reduces the antifungal activity of macrophages. Likewise, exogenous iron enhances and iron chelators inhibit the extracellular growth of P. marneffei. These results suggest that iron availability critically affects i...

A highly discriminatory multilocus microsatellite typing (MLMT) system for Penicillium marneffei.

For eukaryotic pathogens that are depauperate in genetic variation, multilocus microsatellite typing (MLMT) offers an accurate and reproducible method of characterizing genetic diversity; herein we describe the development of an MLMT system for the emerging pathogenic fungus Penicillium marneffei based on 23 microsatellite loci. Screening isolates held within the Centraalbureau voor Schimmelcul...
